Background Information: Granzyme B (GZMB) also known as Cathepsin G-like 1 (CTSGL1), Cytotoxic T-lymphocyte proteinase 2 (Lymphocyte protease), Fragmentin-2, Granzyme-2, Human lymphocyte protein (HLP), SECT, CGL1, CSPB, CTLA1, GRB, which belongs to the peptidase S1 family. Granzyme subfamily GZMB / CTSGL1 contains 1 peptidase S1 domain. The catalytic activity of Granzyme B is “Preferential cleavage: -Asp-|-Xaa- >> -Asn-|-Xaa- > -Met-|-Xaa-, -Ser-|-Xaa-“, and is inactivated by the serine protease inhibitor diisopropyl fluorophosphates. GZMB is necessary for target cell lysis in cell-mediated immune responses. It cleaves after Asp. GZMB seems to be linked to activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution and cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis.
Reconstitution Instructions: Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 µg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 month. For extended storage, it is recommended to store at -20°C.
